Oral magnesium salt — marketed for brain/cognition rather than the muscle-relaxation pitch associated with glycinate.

What people say An oral magnesium salt sold as Magtein, discussed in night stacks and compared with glycinate for sleep or cognition. The brain-focused pitch is not proof of better sleep than glycinate. Doses people talk about
Night-stack elemental discussion~145 mg elemental magnesium orally before bed

Quoted 30–60-minute pre-bed timing, sometimes alongside ~200 mg bisglycinate elemental. Neither number is the weight of Magtein salt.

Clip and capsule-label ambiguity200–400 mg “threonate” in bedtime clips

Usually described as salt in the original discussion, but not verified for every product. A separate NOW Magtein post calls one capsule ~400 mg; its salt-versus-elemental identity is unresolved, not an established maximum.

2026 cognition-trial exposure2 g Magtein salt orally per day; 145 mg elemental magnesium total

One 1 g capsule in the morning and one about two hours before bed for six weeks, with or without food. A selected adult trial, not a community target.

Reported amounts, not a target. Elemental magnesium and magnesium-L-threonate salt mass are different quantities; unclear clip or capsule labels remain unclear.

Timing context & sources

Half-life in the body

No single Magtein wear-off time was established in the checked human evidence.

The cited 2.5-hour figure concerns threonate after calcium L-threonate—not magnesium or a brain effect. Magnesium also occupies regulated tissue stores.

This bounded review does not claim that magnesium has no kinetics. Ligand, circulating magnesium, tissue stores and subjective benefit cannot share one borrowed value.

  • EFSA: magnesium L-threonate assessment (opens in a new tab)2024 sections 3.8.1–3.8.3: dissociation, rat data, and human calcium L-threonate study measuring plasma threonate.Safety/bioavailability assessment, not an efficacy endorsement. Some evidence is applicant-supplied/unpublished; calcium-threonate ligand kinetics are not magnesium brain clearance.
  • NIH: magnesium compartments and regulation (opens in a new tab)Introduction: most magnesium is in bone/soft tissue, less than 1% in serum; kidney regulation.General magnesium physiology, not a measured Magtein half-life or comparison of subjective effects.

Felt duration people report

One person described a few-hour mental boost; another described sleep and next-day effects that later shifted to disrupted sleep.

A rapid sensation, a whole night's sleep and next-day changes are different observations; reports also include overstimulation or no benefit.

These accounts do not establish a common duration, brain-magnesium measurement or product-wide benefit.

  • Lopresti & Smith (2026): six-week Magtein cognition and sleep trial (opens in a new tab)Author/date heading (January 12, 2026); Abstract; sections 2.3–2.4: 100 adults aged 18–45 with dissatisfied sleep, 2 g Magtein/145 mg elemental magnesium daily, one 1 g capsule morning and one two hours before bed; Funding and Conflict of interest.Actual article heading, abstract, participant/intervention methods and funding disclosures inspected. Six-week selected-population study; Threotech funded and supplied product and helped conceptualise the design. Not a glycinate comparison or a universal felt-duration study.

What people say 5

  • Huberman sleep cocktail: 30–60 min before bed with glycinate/theanine/apigenin in the folklore stack. forum
  • r/Biohackers recap of that paper: “Memory sharper, sleep kinda felt better, trackers meh.” That’s the honest read. forum
  • Long-term brain pitch: Why people pay the patent premium vs cheap glycinate. Evidence is still one-ish modern sleep/cognition RCT plus older Magtein papers. forum
  • Cognition (6-week RCT): NIH total cognition composite, working/episodic memory, reaction time, “cognitive age” estimate improved vs placebo on 2 g Magtein. trial
  • Sleep subjective vs objective: Better self-report sleep-related impairment; ring-tracker sleep outcomes no group difference; HR down / HRV up. trial

Doses people talk about 6

  • Huberman-quoted elemental: ~145 mg elemental magnesium from L-threonate, 30–60 min before bed — sometimes paired with ~200 mg bisglycinate elemental. forum
  • Other clip numbers: 200–400 mg “threonate” before bed — usually the salt, not elemental. forum
  • NOW Magtein caps: r/HubermanLab doing 1 cap ~400 mg as “Huberman max” — check whether that’s salt. forum
  • Don’t add 400 mg oxide on top and call it the same experiment. forum
  • Framing: Elemental vs Magtein-salt milligrams get swapped in clips. Read the bottle. forum
  • Lopresti & Smith 2026: 2 g Magtein daily for 6 weeks, supplying 145 mg elemental magnesium/day. The trial used one 1 g capsule in the morning and one about 2 hours before bed, with or without food. trial

How it may feel 5

  • Night 1: Some sleepy, some nothing, some GI. forum
  • Weeks 1–2: Sleep-stack judgment. Glycinate people often already decided. forum
  • If you only wanted muscle-relax sleep: That’s why r/sleep picked glycinate. forum
  • Week 6: The Lopresti & Smith 2026 trial length. trial
  • If sleep trackers don’t move: The paper already said that can happen while cognition scores do. trial

Around the dose 3

  • Glycinate vs threonate is the fork: Posts compare sleep with cognition before combining both; stacking them makes crediting one product difficult. forum
  • Elemental math on the Supplement Facts. forum
  • Sleep trackers may not agree with how you feel: The six-week Lopresti & Smith 2026 paper reported this subjective/objective distinction. trial

Cycles people discuss 3

  • Daily for weeks: The sleep-stack habit is often nightly; the six-week cognition trial split its daily amount between morning and evening. forumtrial
  • Not a 10-day peptide course. forum
  • ~6-week checkpoint: Reassessment at about 6 weeks when cognition is the question echoes the study length; it is not an established stop/restart rule. trial

Timing 3

  • Before bed 30–60 min is the Huberman clock. forum
  • Brain-mag story is days-to-weeks, not a 20-minute benzo. forum
  • With the rest of the night cocktail (glycinate, theanine, apigenin) — confound city. forum

More on what it is 6

  • What it is: Magnesium bound to L-threonic acid. Magtein is the patented salt name on a lot of bottles — industry patent, not a shop SKU. Not a peptide. forum
  • Why people talk: Huberman night stack. r/HubermanLab, r/PeterAttia “is this actually better than glycinate?” forum
  • The fight: Threonate = brain/cognition. Glycinate = chill/sleep/muscles. r/sleep posts skip Huberman’s threonate on purpose for glycinate. forum
  • Dose-label chaos: Huberman clips get quoted as 145 mg *elemental* vs 200–400 mg of *salt* vs 2 g Magtein in the trial. Those are not the same number. forum
  • Not: Not glycinate, not oxide, not IV mag. forum
  • 2026 Lopresti & Smith: 2 g/day Magtein vs placebo × 6 weeks in adults with dissatisfied sleep — better cognition composite / working memory / reaction; subjective sleep-impairment improved; sleep-tracker sleep did not beat placebo as a group. trial

Stacks 3

  • L-theanine / apigenin / glycine: Huberman night kit. forum
  • Magnesium glycinate: Either/or more than plus-plus, in the thoughtful posts. forum
  • Tesofensine formulas sometimes toss threonate in as a cap-mate — different job. forum

Storage notes 2

  • Capsules: Dry bottle. Not a peptide pin. forum
  • No mix instructions here: STUDresearch does not list reconstitution charts. forum

Watch for 6

  • GI / loose stool: “Mag is mag” is community shorthand for gut and loose-stool complaints, not proof that every magnesium form feels the same. forum
  • Sleepy next morning if the night cocktail is heavy. forum
  • Kidney disease / high mag: Mineral, not a candy. forum
  • Patent premium ≠ proven better sleep than glycinate. r/PeterAttia asked that out loud. forum
  • Lopresti & Smith 2026: No significant AE reports in that 6-week cap study — still not a lifetime safety file. trial
  • Research-only framing for this map: Discussion of community practice — not a recommendation to use. forum
