Dimethylglycine (DMG) is a glycine derivative discussed for energy, clearer thinking and methylation. Individual users describe subtle mood benefits or feeling more alert, but also unwanted stimulation and physical symptoms. Hair discussions concern topical products, often combined with caffeine; those findings do not establish benefits from swallowing or injecting DMG.
What DMG is and why people compare it with TMG
- DMG and TMG are different compounds. DMG means N,N-dimethylglycine: glycine with two methyl groups attached to its nitrogen. TMG is trimethylglycine, also called betaine, and DMG is one of its metabolites. Glycine itself and sarcosine are related names, not interchangeable products. Official context Official context
- Methylation interest often starts with a poor TMG experience. Methylation is the transfer of small chemical groups in the body. In MTHFR discussions, people who felt worse with TMG ask whether DMG will behave differently. The discussion offers biochemical speculation rather than a DMG trial, so it cannot establish which genetic profile benefits or that DMG is gentler. Reddit r/MTHFR Reddit r/MTHFR
Benefits people describe
- More energy and easier conversation, in one continuing account. A poster described clearer thinking and more energy after a few days, then better mood and easier conversation. Their later update credited vitamin B injections and dietary changes more strongly. The early improvement remains a report, but the follow-up weakens any claim of a lasting DMG-specific effect. Personal report
- One person described a subtle mood change. One sublingual user described a small, steady smile and said TMG had done less for them. They supplied no amount or answer to a question about consistency. That is a useful personal comparison, not evidence that sublingual DMG reliably improves mood. Personal report
Doses people discuss
- 125 mg three times daily, or 500 mg each morning, in supplement accounts. The first totals 375 mg/day and comes from the mouth/eye complaint report. The 500 mg morning account described energy and clarity. Neither specifies swallowed versus sublingual use. These are individual reports, not an established common range or a recommended ladder. Personal report Personal report
- Another account split 500 mg/day into five 100 mg amounts. That person was already using several methylation supplements. A separate Reddit question mentions products sold at 125 mg per dose but gives no DMG outcome. The sublingual mood report gives no amount; none of these numbers supplies an injection or shampoo dose. Half-life
- No usable DMG half-life estimate in the inspected community accounts. The methylation-stack poster asked about half-life without establishing a number. Divided doses and felt effects do not measure clearance. Personal report
Onset: when people notice a change
- A few days for one energy report; 2–3 days for mouth symptoms. These describe different outcomes during repeated supplement use. The sublingual smiling report has no clock. There is no dependable minutes-to-hours onset window here. Personal report Personal report Personal report
Duration: how long a change lasts
- Single-dose benefit duration remains unclear. A steady smile has no stated hour count. The energy account describes successive days, and its later improvement had other explanations. Neither report establishes all-day coverage, a withdrawal syndrome or how long benefits persist after stopping. Personal report Personal report
Cycles and time off
- Several weeks of use is reported; a standard on/off cycle is not. The methylation-stack account continued across weeks. The shampoo user described a few months of use. Neither supplies a planned break schedule or shows that cycling restores sensitivity; a supplement course and repeated topical use also answer different questions. Good to know
- More stimulation was not always comfortable. The methylation-stack poster described feeling revved up and later unusual urine odor. They felt worse after reducing DMG, but other supplements and later antibiotics complicate that story. It does not demonstrate dependence, detoxification or an immune-system reset. Personal report
- Mouth and eye symptoms were reported. One person reported mouth bumps, worsened eye soreness and eyelid twitching after starting DMG. They already had blepharitis and mentioned a possible dental problem. Replies proposed nutrient deficiencies, but neither that explanation nor the poster’s detox theory was established. Personal report
- A natural metabolite can still have uncertain supplement effects. The small oral trial found similar self-reported tolerability to placebo, and the shampoo trial reported no adverse events during follow-up. Those findings cannot establish safety for every amount, long-term use, combinations, pregnancy or injection. Community reports do not establish safety or efficacy. Official context Official context
What the human studies can and cannot answer
- A small oral trial did not confirm the fatigue benefit. In 30 people with progressive multiple sclerosis, 125 mg/day of oral DMG for 12 months did not significantly improve fatigue, cognition, walking or disability versus placebo. This is disease-specific research, not a community dose recommendation or proof that every user will feel nothing. Official context
- The shampoo result belongs to a combination. A 24-week trial in 154 men compared shampoo containing 1% DMG sodium salt plus 1% caffeine with a base shampoo. Hair-pull results favored the active product. The manufacturer funded it, company employees coauthored it, and only 30 participants had imaging measurements; there was no DMG-only arm. Official context
- Supplement reports, shampoo results and supplier blends cannot be swapped. The shampoo trial does not establish hair growth from oral DMG, and neither that trial nor supplement anecdotes validates injected DMG or a multi-ingredient supplier blend. Likewise, the betaine pharmacokinetic paper measured DMG after giving TMG; its betaine half-life is not a half-life for a DMG supplement. Official context Personal report Official context
What community discussion adds
- The methylation explanation remains a hypothesis in these discussions. The MTHFR question asks whether DMG resembles TMG or NAC. Its respondent explicitly had not tried DMG and inferred benefits from a pathway diagram and shopping reviews. That distinction matters: an appealing mechanism is not another firsthand success, and a bad TMG reaction does not predict a good DMG reaction. Reddit r/MTHFR
- Stack changes can change the interpretation of an early win. The energy poster’s later explanation centered on vitamin B injections and diet. The revved-up poster was using TMG, B12 forms, methylfolate and other supplements, with antibiotics added later. These histories are more informative than attributing every later improvement to DMG alone. Personal report Personal report
- Less visible shedding and regrowth are different outcomes. A shampoo user reported a few fewer hairs in their hand after several months but no regrowth. Others questioned the company-led evidence and asked for updates; no later outcome from that user was visible. This mixed account concerns the finished shampoo, not isolated DMG.
