Community talk. May be wrong. Not medical advice. Not a protocol. Not for human or animal use.
Systemic oral experimental small molecule (curcumin-derived neurotrophic).
The author described increasing from 10 to 40 mg and later about a year of use; fisetin and other factors entered the account.
One commenter described once-morning use and stopped because of possible sleep impact; another author reported 35–100 mg/day in divided use with several stacks.
Produced the 1.5-hour plasma and 2.5-hour brain half-life estimates; not a human-equivalent amount or efficacy schedule.

Timing context & sources
Half-life in the body
After 20 mg/kg PO in mice, J147 half-life was reported as 1.5 hours in plasma and 2.5 hours in brain.
The same study reported 28% oral bioavailability and brain concentration at two hours. The two-hour concentration was not labeled Tmax.
Mouse-only single-dose PK; no public human Phase 1 results; the range combines two compartments and cannot be transferred to consumer products.

- ClinicalTrials.gov NCT03838185 — J147 single-ascending-dose Phase 1 (opens in a new tab)Registry record: randomized double-blind placebo-controlled single-ascending oral-dose study; 64 actual participants; start 2019-01-22 and completion 2020-02-01; no posted results.Registration record only; no public dose table, PK curve, adverse-event results or publication was available in the reviewed record.
Felt duration people report
One long-running author reported about four days to onset and about four days to disappearance after stopping; other users reported sedation, sleep effects or little benefit.
The same author described roughly a year of continued benefit and substantial co-use. Reddit reports used 30–100 mg/day and included bromantane, phenibut, alcohol or other nootropics.
Self-report, no product verification or washout, psychiatric indications and medications, multiple co-exposures and no objective cognitive testing.
- LongeCity — J-147 and fisetin self-experiment thread (opens in a new tab)Topic-starter posts describe 10 mg increases to 40 mg, roughly four-day onset/offset, nearly a year of claimed benefit, sleepiness/cold, early energy, blood-pressure reports, weight loss, nonresponders and later fisetin/metformin/berberine context.Uncontrolled psychiatric self-experiment, unverifiable synthesis, extensive co-use and second-hand reports; vendor and sourcing statements are not reproduced.
- r/NooTopics — J147 experience and follow-ups (opens in a new tab)Original and replies describe mild solo mood effects, alcohol/phenibut/bromantane confounding, 35–100 mg/day split use, separate 30–45 mg morning use, sleep impact, early naps, hunger and marked fatigue.Multiple anonymous users, shifting amounts and stacks, no identity testing, clinical assessment or objective outcomes.
What people say
- n=1 “cleaner focus”: Thin. A few LongeCity/BioMind logs say words come easier or afternoon crash is quieter — confounded by Dihexa, sleep, and expectancy. anecdote
- Why it sits next to Dihexa: One is HGF/c-Met peptide lore, one is mitochondrial small-molecule lore. Clinics sell the pair as a brain stack, not a proven combo trial. forum
- Honest empty cell: There is no dense r/Nootropics album the way racetams or kratom have. Most “benefits” are paper titles. forum
- Mouse memory / AD models: Salk papers: memory rescue, less AD pathology, “younger” mitochondria in old mice. That is the citation, not a human before/after. animal
- Cell neurotrophic screens: Nanomolar protection in HT22 / cortical neuron assays is why vendors call it potent. lab
Doses people talk about
- Community oral amounts: Vendor lists often show 10–20 mg/day, while inspected self-reports described 10 mg increased to 40 mg, 30–45 mg each morning, and 35–100 mg/day in divided use. None is a validated human dose. forumanecdote
- Lower n=1: 5–10 mg “in case it’s strong.” No titration science behind it. forum
- Not curcumin grams. This is a different molecule at milligrams, not turmeric scoops. forum
- Framing: Vendor milligrams and mouse mg/kg — not a labeled dose. Human PK is not a public label. forum
- Mouse work: ~20 mg/kg oral in the PK paper people cite. Do not cargo-cult that onto a 70 kg human. animal
How it may feel
- Days 1–7: Reports conflict. One long-running author described positive effects starting around day 4, with early excess energy or midday sleepiness/cold; Reddit users reported mild mood effects, naps, hunger or marked fatigue, while others noticed little. forumanecdote
- Days to months: One author placed onset and loss of claimed mood, energy and cognition effects about four days after starting or stopping and reported persistence during roughly a year of use. Other accounts describe sleep disruption, fading early energy or no clear cognitive effect. forumanecdote
- Stop: No withdrawal story in the thin logs. forum
- Months: No human time-course. Mouse work is weeks to months of daily dosing. animal
Cycles people discuss
- Daily while the bottle lasts: 4–8 week experiments, then people keep it in the stack or forget it. forum
- Not a racetam cycle. No choline-dump ritual. forum
- Stacked continuously in “brain boxes” rather than 5-days-on. forum
- Stop if headache / overstim is the only practical rule in the logs. forum
Timing
- Once-daily oral is the vendor ritual. forum
- Morning with the rest of the nootropic stack so they can notice overstim. forum
- Mouse oral PK: After one 20 mg/kg PO dose, the reported half-life was 1.5 hours in plasma and 2.5 hours in brain, with 28% oral bioavailability; brain concentration at 2 hours was a sampling result, not the reported Tmax. Human Phase 1 PK results are not posted. animaltrial
- Metabolic instability talk in some vendor chem sheets — people still swallow a daily cap. lab
More on what it is
- Why people talk about it: BioLongevity-style “BioMind” boxes stack it with Dihexa. LongeCity / r/Nootropics treat it as the mitochondrial AD pill you can mail-order. forum
- Not Dihexa, not racetams, not methylene blue. Different stack drawer that gets thrown in the same “brain in a box.” forum
- What it is: A Salk/Cadet-lab curcumin analog (Schubert) built as an Alzheimer’s candidate. Oral. Still not an approved human drug in these threads. Not a peptide. trial
- How it works (plain): Later Salk work tied J147 to mitochondrial ATP synthase / aging-clock talk in mice — not “more curcumin.” The Instagram version is “reverses Alzheimer’s in mice.” trial
- Mouse PK people quote: ~20 mg/kg in mice produced brain levels well above cell EC50 at 2 hours. That is not a 10 mg capsule conversion. animal
- Human status: A randomized, placebo-controlled single-ascending-dose Phase 1 enrolled 64 healthy young and older volunteers and completed in February 2020. The registry still posts no results, so no human PK curve, adverse-event table or dose conclusion is publicly available there. trial
Stacks
- + Dihexa: The BioMind-class pair. Two thin human diaries multiplied. forum
- + Semax / Selank / P21: Same cognitive-peptide drawer. forum
- Solo first is rare — most logs are polypharmacy. forum
- Not with a new stimulant on day one if they want to know which thing is the headache. forum
Storage notes
- Capsules or dry powder. Cool, dry, light-tight. forum
- Not BAC water. If a “nasal J-147” listing appears, treat it as a different unverified product. forum
Watch for
- Reported experience range: Sleepiness, feeling cold, hunger, marked fatigue, early excess energy, sleep disruption, anxiety, raised blood pressure and unintended weight loss all appear in small self-report threads, alongside mild benefit or no clear effect. Product identity, psychiatric treatment, alcohol and other nootropics confound attribution. forumanecdote
- Identity / dose: 10 mg gray caps are not Salk GMP. forum
- Do not convert 20 mg/kg mouse to a human gram. forum
- Cancer-pathway / growth-signal stacking lore when piled with Dihexa and other neurotrophics — forum caution, not a trial. forum
- Human data is the caution. Mouse AD rescue ≠ a supplement. animal
- Not medical-grade Alzheimer’s care. trial
