NSI-189 is an experimental small molecule people discuss for emotional numbness, loss of pleasure and foggy thinking. Some describe feeling engaged again; others report little benefit, anxiety, fatigue or worse cognition. Stronger emotion can feel welcome or become difficult to manage.

- Feeling something again. In the 2022 poll thread, one commenter welcomed crying at a show after a month. Another remembered increased emotionality but no permanent change. These accounts describe emotional responsiveness, not necessarily relief from depression. Reddit

These are self-reported amounts, not instructions or a safe range. mg means milligrams. Form, route and daily total matter.
- Oral phosphate: 40 mg total per day in one month-long account. That author did not get the hoped-for acute benefit. They also mentioned 80 mg experiments without a clear frequency, then a sublingual trial; those details do not establish an effective range. Reddit
- Sublingual phosphate: roughly 5–7 mg twice daily in another account. That implies about 10–14 mg per day, but the author estimated use from how long the powder lasted rather than weighing each dose. - Oral freebase: 80–110 mg each morning in one positive log. The author reached this after little response to initial 40 mg sublingual use, then a slight lift with 40 mg swallowed. How long changes feel noticeable

Cautions that matter
- Greater emotional intensity can become destabilizing. One long-use contributor described impulsivity, irritability and difficulty functioning. Another reported psychosis and persistent tingling after prolonged use with several other compounds. The accounts are serious but cannot isolate NSI-189 as the cause. Reddit
- Cognitive worsening and fatigue are part of the record. A 2024 commenter recalled months of brain fog and poor short-term memory after about two weeks of use. - Combining substances makes the story harder to interpret. NAC, caffeine, other nootropics and psychiatric medicines appear across accounts. What the human research adds
- A small early trial measured oral-phosphate clearance. Fava and colleagues studied 24 people with major depression for 28 days, using oral phosphate at 40 mg once, twice or three times daily (40, 80 or 120 mg/day). Neuralstem’s 220-person phase 2 study missed its primary depression endpoint at both tested amounts, despite improvements on some self-rated and attention/memory measures, but not Cogstate testing.
